Ubiquitination is a reversible post-translational modification that is involved in almost all biological processes, including proteome homeostasis, DNA damage repair, gene transcription, protein endocytosis and trafficking, stem cell pluripotency. Ubiquitination is catalysed by a three-enzyme E1-E2-E3 cascade, while the ubiquitin tags are deconjugated from substrate proteins by deubiquitinases. My laboratory at the Structural Genomics Consortium, University of Toronto, utilizes structural biology and biophysical techniques to study the structure, function, and regulation of ubiquitin regulatory enzymes of human disease relevance at molecular level. We are also developing inhibitors to regulate the activity of these enzymes for target validation.
